The Pex7 (peroxisomal biogenesis factor 7) gene encodes a member of the peroxisomal assembly (PEX) proteins and is involved in the transport of enzymes for the assembly and function of peroxisomes. Mutations in this gene are associated with peroxisome biogenesis disorders (PBDs). Murine Pex7 expresses two transcripts.&nbsp; The larger transcript -201 generates a 318aa protein.&nbsp; Transcript -203 generates a smaller 292aa protein.&nbsp; 203 has a smaller 3&rsquo; UTR but starts at the same codon in exon 1 as 201. -203 is missing exon 4 which encodes 26 amino acids. Loss of exon 3 would impact both isoforms. To create the conditional Pex7em2Lutzy allele, CRISPR/Cas9 endonuclease-mediated genome editing was used to insert loxP sites flanking exon 3. Prior to Cre recombinase exposure, mice heterozygous or homozygous for this floxed allele are viable and fertile, with no reported phenotypic abnormalities. Upon exposure to Cre recombinase, deletion of the floxed sequences results in a null allele.
