TYW3 transgenic mice express yellow fluorescent protein under the direction of the neuron-specific mouse Thy1 gene promoter. While the transgenic insert contained other reporter genes, neither the WGA (wheat germ agglutinin) nor lacZ expression is detectable. YFP is flanked by loxP sites, and can therefore be deleted by Cre recombinase. YFP is detected in 6 types of retinal ganglion cells (RGCs), all of which share a dendritic lamination pattern: dense dendritic arbors are primarily localized to central third (S3) of the inner plexiform layer. The transgenic insertion site was mapped (using Targeted Locus Amplification) to Chromosome 1, approximately 7kb upstream of the initiation codon of the Khdrbs2 gene. The transgenic insertion resulted in a 500kb deletion upstream of the insertion site. Khdrbs2 mRNA levels are reduced by approximately 45% in TYW3 homozygotes, compared to wildtype controls. Quantitative fluorescent PCR analysis reveals 5 copies of the transgene were inserted at the single genomic site. The Khdrbs2 gene encodes an RNA-binding protein that regulates alternative splicing. When exposed to Cre recombinase the YFP cassette is excised. Mice that are homozygous for the transgene are viable and fertile.
