ProcrCreERT2-IRES-tdTomato mice have an inducible cre/ERT2 fusion gene followed by a tdTomato reporter inserted into the start codon of the protein C receptor (Procr) gene. Insertion of this fusion gene abolishes expression of PROCR. Procr encodes an N-glycosylated type I membrane protein involved in activation of protein C. Mice that are heterozygous for this knock-in/knock-out allele are viable and fertile; homozygous embryos die by E10.5.When crossed with a strain containing a loxP site flanked sequence of interest, the offspring are useful for generating tamoxifen-induced, Cre-mediated targeted deletions. Tamoxifen administration induces Cre recombination in the basal layer of the mammary epithelium and recapitulates endogenous Procr expression. Procr+ cells mark a unique population of multipotent mammary stem cells (MaSCs), are localized to the basal layer, and exhibit characteristics of epithelial to mesenchymal transition (EMT). Inducible Cre recombinase activity is detected in the basal cells (3-7%) and in the stromal cell department. The donating investigator notes that tdTomato proteins forms aggregated punctates, and may not be sufficiently bright for detection in some tissue.

ProcrCreERT2-IRES-tdTomato are knockin/knockout mice with an inducible cre/ERT2 fusion gene followed by a tdTomato reporter inserted into the ATG codon of the protein C receptor (Procr). These mice can be used to identify multipotent mammary stem cells (MaSCs) in the basal layer of the mammary epithelium.
