Mice harboring the vil-Cre transgene have the 9-kb mouse, Vil1, villin 1, promoter directing expression of Cre recombinase to the intestinal epithelia and kidney, and may be useful for generating conditional mutations in cells of the crypt villus axis and kidney. This strain also is available on the C57BL/6J background (Stock No. 035595).

These transgenic mice express Cre recombinase under the direction of the 9-kb mouse Vil1, villin 1, promoter. When crossed with a strain containing loxP site flanked sequence of interest, Cre-mediated recombination results in tissue-specific deletion of the target. Recombination occurs in the crypt villus axis of the intestinal epithelia (enterocytes, goblet cells, and putative undifferentiated progenitor cells in the crypts) and kidney. No transgene expression or Cre recombinase activity was detected in non-epithelial intestinal cells, spleen, skeletal muscle, liver or lung. Cre recombinase activity is detected in the visceral endoderm and developing gut of E10.5 embryos. The Donating Investigator has not attempted to make this strain homozygous. Hemizygotes are viable and fertile. Homozygous viability/fertility has not been tested (January 2019).

A transgenic construct containing sequence encoding cre, Cre recombinase, followed by an SV40 polyadenylation signal, under the control of the 9 kb mouse Vil1, villin 1, gene promoter, was injected into fertilized B6D2 (C57BL/6 x DBA/2) mouse eggs. Founder line 20 (Fo20) was subsequently established. Southern blot analysis revealed a single copy insertion of the transgene. The mice were then backcrossed to C57BL/6NCrl for 10 generations. During backcrossing, the Y chromosome may not have been fixed to the C57BL/6NCrl genetic background. Cryopreserved sperm was sent to The Jackson Laboratory Repository. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ (Stock No. 005304) oocytes.
