Description

Cbln2^flox mice carry a targeted conditional reporter of Cbln2 (cerebellin 2 precursor protein) activity. Any cell that expresses Cbln2 will also express IRES-mVenus.  Following cre-mediated recombination, the IRES-mVenus is deleted and the cells will express IRES-tdTomato.

Detailed description

Cerebellins are secreted glycoproteins that belong to the C1q and tumor necrosis factor (TNF) superfamily. They function as trans-synaptic cell-adhesion molecules that critically contribute to the formation and/or function of synapses and are differentially expressed in selective subsets of neurons throughout the brain.

These Cbln2^flox mice carry a targeted conditional reporter of Cbln2 (cerebellin 2 precursor protein) activity. Any cell that expresses Cbln2 will also express IRES-mVenus. Following Cre-mediated recombination, the IRES-mVenus is deleted and the cells will express IRES-tdTomato. Both mVenus an tdTomato can be visualized directly, but the tdTomato is significantly weaker than the mVenus, thus the use of antibody for detection is recommended.

Cbln2 and Cbln4 (see stock# 032950) are broadly expressed in largely non-overlapping subpopulations of excitatory cortical neurons, but only sparse expression was observed in excitatory hippocampal neurons of the CA1- or CA3-region. Similarly, Cbln2 and Cbln4 are selectively expressed, respectively, in inhibitory interneurons and and excitatory mitral projection neurons of the main olfactory bulb; here, these two classes of neurons form dendrodendritic reciprocal synapses with each other. A few brain regions, such as the nucleus of the lateral olfactory tract, exhibit astoundingly high Cbln2 expression levels.

Development

A loxP site was introduced to intron 1 of mouse Cbln2 genomic sequences, and IRES-mVenus, an FRT-flanked neomycin resistance cassette, and a loxP-SA-Stop-IRES-tdTomato-WPRE-SV40-pA cassette were placed in intron 3 via homologous recombination in (129X1/SvJ x 129S1/Sv)F1-Kitl⁺-derived R1 embryonic stem (ES) cells. The neomycin cassette was excised from resultant mice through crosses with animals expressing Pgk1-FlpO recombinase (Stock# 011065). This strain was backcrossed to C57BL/6J at least two times by the donating laboratory.

Molecular note: A loxP site was inserted upstream of exon 2 and an IRES-mVenus reporter, FRT flanked neo cassette loxP site, stop cassette and IRES-tdTomato reporter were inserted downstream of exon 2 via homologous recombination. Flp mediated recombination removed the neo cassette.
