Description

HRGP-cre mice express Cre recombinase under the control of the retinal cone receptor cell-specific, human OPN1LW, opsin 1, long wave sensitive gene promoter. They are suitable for Cre-lox studies of cone photoreceptors.

Detailed description

These HRGP-cre transgenic mice express Cre recombinase under the direction of the human OPN1LW, opsin 1, long wave sensitive (also referred to as: human red/green pigment, HRGP), gene promoter. When crossed to a beta galactosidase-expressing reporter strain (R26R reporter mice), Cre recombinase activity was detected in cone photoreceptors of the retina. Transgene expression in adult transgenic mice does not alter cone photoreceptor cell distribution and electroretinography (ERG) characteristics. Mice that are hemizygous for the transgene are viable and fertile. The Donating Investigator expects homozygotes to be viable and fertile. Homozygous viability/fertility has not been tested (January 2019).

Development

A transgenic construct containing sequence encoding cre recombinase under the control of the human OPN1LW , opsin 1, long wave sensitive (also referred to as: human red/green pigment, HRGP) gene promoter (which contains 5' upstream sequence and part of the red pigment gene), followed by a mouse mouse metallothionein (Mt1) polyadenylation signal sequence, was injected into fertilized FVB/N mouse eggs. Founder line 4 was subsequently established. The mice were then backcrossed occasionally to FVB wildtype mice by the donating lab (see SNP note below). Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize FVB/NJ oocytes (Stock No. 001800).



In 2019 a 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. Most of the 43 markers throughout the genome were segregating with an unknown source. These data suggest the mice sent to The Jackson Laboratory Repository were on a mixed genetic background.
