Description

Pthrp-creER mice express cre/ERT2 fusion gene from the parathyroid hormone-like peptide (Pthlh) promoter. These mice may be useful for lineage mapping skeletal stem cell lineages.

Detailed description

Mice hemizygous for the BAC Pthrp-creER transgene express a cre/ERT2 fusion gene from the parathyroid hormone-like peptide (Pthlh) promoter (otherwise known as parathyroid hormone-like hormone, Pthrp). PTHrP encodes a cytokine that regulates endochondral bone development and epithelial-mesenchymal interactions during the formation of the mammary glands and teeth. Homozygous mice are viable and fertile. Restricted to the cytoplasm, cre-ERT2 can only gain access to the nuclear compartment after exposure to tamoxifen. When Pthrp-creER mice are bred with mice containing loxP-flanked sequences, tamoxifen-inducible Cre-mediated recombination will result in deletion of the floxed sequences in the Cre-expressing cells of the offspring. With this line (Line909) Cre recombination occurs specifically in chondrocytes of the resting zone of the growth plate after tamoxifen administration at postnatal day 6.

When bred with Rosa26 reporter strains (such as Ai14 (Stock No. 007914) and Ai6 (Stock No. 007906), Pthrp-expressing cells can be permanently marked upon tamoxifen injection at postnatal day 6.

Of note, the donating investigator states that Pthrp-creER does not induce recombination efficiently when bred with some Rosa26 reporter strains (such as R26R-Confetti (Stock No. 013731) and R26R-EYFP (Stock No. 006148), therefore recombination events need to be validated on an individual target allele basis. In addition, tamoxifen injection after postnal day 6 does not efficiently mark Pthrp-expressing cells in the resting zone of the growth plate

Development

Bacterial artificial chromosome (BAC) library (CHORI) was used to obtain a BAC (#RP23-27F7) containing the entire mouse parathyroid hormone-like peptide (Pthlh) gene. This BAC was modified to replace the native start codon of the Pthlh gene with a cre/ERT2 fusion gene (Cre recombinase fused to a human estrogen receptor ligand binding domain), followed by an SV40 polyadenylation sequence. This modified BAC was microinjected into fertilized B6SJLF1 oocytes. Pthrp-creER mice from founder line 909, with a single copy of the transgene, was established and then subsequently maintained by breeding transgenic mice with C57BL/6J inbred mice for at least 11 generations. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J (Stock No. 000664) oocytes.
