Detailed description

CRISPR/cas9 endonuclease mediated genome editing (NHEJ) of the Gdpd3, glycerophosphodiester phosphodiesterase domain containing 3, gene was used to delete exons 2 through 7. The targeted Gdpd3 gene encodes a lysophospholipase. Homozygous mice are viable and fertile. No gene product (mRNA) in liver, lung, and spleen tissue is detected by qPCR analysis. Heterozygotes express half of the wildtype level of mRNA in liver, lung, and spleen tissue. Homozygotes are viable and fertile.

Development

CRISPR/cas9 endonuclease mediated NHEJ genome editing of the Gdpd3, glycerophosphodiester phosphodiesterase domain containing 3, gene was used delete exons 2 through 7, in C57BL/6J fertilized eggs. Correctly targeted embryos were transferred to pseudopregnant females. Correctly targeted pups were identified by sequencing and PCR. The mice were crossed to C57BL/6J mice for at least 1 generation.  Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
