When these Hip1 knock-out mice are bred to a cre recombinase expressing strain, the resulting double mutant mice express a single copy of human HIP1 in cre-expressing tissue(s). The targeted Hip1 gene encodes a protein that is involved in endocytosis and interacts with huntingtin protein. Expression of HIP1 chromosomal translocation complexes are associated with leukemia and non-small cell lung cancer. HIP1 expression is also associated with prostate and colon cancers. Homozygotes are viable but homozygous males are not fertile. Onset of phenotype occurs at 4 months of age. By 6 months of age, HipLSL homozygotes exhibit reduced body weight, generalized weakness and degenerative spinal defects (kypholordosis). Phosphocholine levels are decreased in kidney, lung, liver and spleen tissues but not brain. EGFP expression is detected in peripheral blood of homozygous animals.

HipLSL mutant mice have a loxP-flanked STOP sequence followed by the human HIP1 (huntingtin interacting protein 1) cDNA and IRES-GFP, followed by a polyadenylation signal sequence, in intron 1 of the Hip1 gene. When bred to mice expressing Cre recombinase, the STOP sequence is deleted and the human HIP1 and EGFP expression is observed in the cre-expressing tissue(s) of the double mutant offspring. These mutant mice may be useful in studies of choline metabolism and its role in cancer development, as well as spinal homeostasis.
