Description

Brca2^flox mice have loxP sites flanking exon 11 of the Brca2 gene. This strain may be useful for generating conditional mutations in applications related to breast cancer.

Detailed description

The targeted Brca2 gene encodes a tumor suppressor protein that is involved in DNA repair. Mutations in this gene have been associated with familial breast-ovarian cancer susceptibility, prostate cancer, Fanconi anemia and Wilms tumor. Brca2^flox mice have loxP sites flanking exon 11 of the breast cancer 2, early onset (Brca2) gene. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific cre recombinase resulting offspring will have exon 11 removed in the cre-expressing tissues of the offspring. Removal of the floxed sequence creates a null allele.

Development

A targeting vector designed to insert a loxP-flanked neo cassette in intron 10, and a third loxP site downstream of exon 11 of the breast cancer 2, early onset (Brca2) gene. The construct was electroporated into 129P2/OlaHsd-derived E14IB10 embryonic stem (ES) cells. Correctly targeted ES cells were transiently transfected with a Cre expression plasmid to delete the neo cassette. Resulting ES cells contained multiple gene rearrangments; intact floxed-exon 11, intact floxed-neo cassette, or excision of both exon 11 and the neo cassette. Correctly targeted ES cells, containing only the floxed-exon 11, were injected into C57BL/6 blastocysts and resulting chimeric mice were bred with FVB/N mice. This line of Brca2^flox mice were maintained with other alleles before this allele was separated out, creating mice sustained as this single allelic strain.

Marker symbol: Brca2
Marker name: breast cancer 2, early onset
Marker synonyms: BRCC2; BROVCA2; FACD; FAD; FAD1; FANCD; Fancd1; GLM3; PNCA2; RAB163; XRCC11
