Development

A targeting vector designed to insert a loxP-flanked neo cassette in intron 10, and a third loxP site downstream of exon 11 of the breast cancer 1, early onset (Brca1) gene. The construct was electroporated into 129S6/SvEvTac-derived TC1 embryonic stem (ES) cells. Correctly targeted ES cells were transiently transfected with a Cre expression plasmid to delete the neo cassette. Resulting ES cells contained multiple gene rearrangments; intact floxed-exon 11, intact floxed-neo cassette, or excision of both exon 11 and the neo cassette. Correctly targeted ES cells, containing only the floxed-exon 11, were injected into blastocysts and resulting chimeric males were bred with Black Swiss females. This line of Brca1^flox mice was maintained with other alleles before this allele was separated out, creating mice sustained as this single allelic strain.
