This Casp1Δ10 knock-out allele has a 10 bp deletion in exon 8 of the mouse Casp1 gene. This strain is useful in studies of inflammasome processes.

Detailed description

Casp1 (caspase 1) is essential for NLRC4 (NLR family, CARD domain containing 4)-induced pyroptosis and cytokine processing in macrophages. In intestinal epithelial cells, NLRC4-dependent IL18 release also requires Casp1.

These Casp1Δ10 mice carry a CRISPR/Cas9-generated 10 bp deletion in exon 8 of the Casp1 (caspase 1) gene that creates a knock-out allele.

Development

This CRISPR/Cas9-generated mutation was created by injecting Cas9 and sgRNA AGTCCCCGAGTGAAAAGTA into B6(Cg)-Tyr^c-2J/J embryos. This resulted in a 10 bp deletion (AGGGGCTCAC) in exon 8 of the Casp1 gene, knocking out its expression. This line was backcrossed to C57BL/6J for 4 generations to remove the Tyr^c-2J albinism allele and off-target CRISPR mutations.

The mice do not carry the loss-of-function mutation in Casp4 (Casp11) that is present in Casp1 knockout mice made on the 129S2 genetic background (see Stock No. 032661).
