Ly6aCre knock-in/knock-out mice express cre recombinase and nuclear-localized EGFP from the mouse Ly6a (Sca1) promoter in Ly6a-expressing cells, including cardiac vascular endothelial cells.

Cardiac cells expressing Ly6a (lymphocyte antigen 6 complex, locus A; also called Sca1) predominantly contribute to the cardiac vasculature, particularly in the endothelial cell expansion and remodeling that occurs early after birth or following myocardial infarction (MI).
These Ly6aCre knock-in/knock-out mice express cre recombinase from the mouse Ly6a promoter in Ly6a-expressing cells, including cardiac vascular endothelial cells. Expression of Ly6a is eliminated by the targeted allele. Though present in the targeting construct, IRES-driven expression of nuclear localized EGFP is not observed. Heterozygotes and homozygotes are viable and fertile with no overt developmental or adult phenotype. Expression of a cre-dependent TdTomato reporter has been confirmed in the heart and kidney via immunohistochemistry.
Crosses with cre-responsive R26-TdT reporter mice (see Stock No. 007914) enable permanent labeling and lineage tracing of cells that normally express Ly6a throughout development and postnatal growth, or after cardiac injury. Though rare cardiomyoctes are labeled, this may be the result of fusion events from lineage-traced leukocytes. Approximately 30% of total bone marrow cells are labeled (mostly mature hematopoietic cells).
A cDNA encoding cre-IRES-EGFPnls (nuclear localized EGFP) as well as an FRT-flanked neomycin cassette was recombined in-frame with the Ly6a ATG start codon (exon 2) via homologous recombination in 129S6/SvEvTac-derived W4 embryonic stem (ES) cells. Resultant mice were crossed with Rosa26-Flpe mice (Stock No. 009086) to delete the neomycin cassette. This strain was backcrossed to C57BL/6J for 10 generations by the donating laboratory.
Though present in the targeting construct, IRES-driven expression of EGFPnls is not observed.

In case of dissatisfaction for a valid reason and claimed in writing by a purchaser within ninety (90) days of receipt of, PRODUCTS or SERVICES, JACKSON will, at its option, provide credit or replacement for the PRODUCT received or the SERVICES provided; JACKSON makes no other representations and this shall be the exclusive remedy of the purchaser. Consistent with the requirement for a written understanding regarding animal care and use, the JACKSON Animal Care and Use Committee will review the animal care and use protocol(s) associated with any SERVICES to be performed at JACKSON, and JACKSON shall have ultimate responsibility and authority for the care of animals while on site or in JACKSON custody.
In no event shall JACKSON, its trustees, directors, officers, employees, and affiliates be liable for any causes of action or damages, including any direct, indirect, special, or consequential damages, arising out of the provision of MICE, PRODUCTS, or SERVICES, including economic damage or injury to property and lost profits, and including any damage arising from acts or negligence on the part of JACKSON, its agents or employees. Unless prohibited by law, in purchasing or receiving MICE, PRODUCTS, or SERVICES from JACKSON, purchaser or recipient, or any party claiming by or through them, expressly releases and discharges JACKSON from all such causes of action or damages, and further agrees to defend and indemnify JACKSON from any costs or damages arising out of any third party claims.
MICE, PRODUCTS or SERVICES are to be used in a safe manner and in accordance with all applicable governmental rules and regulations.
