K8.tGPD mice co-express a fast-maturing short half-life (2 hours) turbo green fluorescent protein (tGFP) and an inducible suicide gene, the monkey diphtheria toxin receptor from the mouse keratin 8 promoter. K8 is a type II keratin that forms an intermediate filament in simple single-layered epithelial cells. In the mammary gland, K8 is restricted to the luminal compartment of mammary gland epithelia. In lung, GFP expression restricted to K8-positive cells. Mice hemizygous for the transgene are viable and fertile. The suicide gene, DTR or HBEGF, is induced by administration of diphtheria toxin and results in a lethal response within 48 hours. This mutant mouse strain may be useful in monitoring transitions between differentiation states and in eliminating specific cell types in vivo and in vitro.
