Description

K14.tRPT mice co-express a fast-maturing destabilized red (orange) fluorescent protein (tRFP) and an inducible suicide gene, thymidine kinase, in the basal compartment of mammary gland epithelia. This mutant mouse strain may be useful in monitoring transitions between differentiation states and in eliminating specific cell types in vivo and in vitro.

Detailed description

K14.tRPT mice co-express a fast-maturing destabilized red (orange) fluorescent protein (tRFP) and an inducible suicide gene, herpes simplex virus (HSV) thymidine kinase (TK) from the human keratin 14 promoter. K14 directs expression to the basal compartment of mammary gland epithelia. Females hemizygous (homozygous?) for the transgene are viable and fertile. The donating investigator reports that males carrying the transgene are “nearly” sterile. The suicide gene, TK, is induced by administration of ganciclovir (GCV). Both high and low doses of GCV result in massive weight loss or a lethal response. GCV-treated mammary gland develops a loss of mammary ductal organization and increased lymphocyte infiltration. This mutant mouse strain may be useful in monitoring transitions between differentiation states and in eliminating specific cell types in vivo and in vitro.

Of note, a similar strain, FVB/N-Tg(Krt8-GFP,-HBEGF)#Muth/J (Stock No. 032555) co-expresses a turboGFP and an simian diphtheria toxin receptor in the luminal compartment of mammary gland epithelia as well as other K8 expressing cells.

Development

The transgenic construct was designed with a human keratin 14 promoter driving co-expression of both a turbo red fluorescent protein (tRFP) and a suicide gene, herpes simplex virus (HSV) thymidine kinase (TK), separated by a P2A peptide plus a neomycin selection cassette. The donating investigator reports that the neomycin cassette was removed. The presence of the P2A peptide allows translation-coupled cleavage of the fluorescent protein and TK. tRFP is a fast-maturing destabilized red (orange) fluorescent protein. Founder line 4264 was bred to FVB/N mice.
