Description

These Osr1-BF transgenic mice are a useful tool for the isolation and purification of OSR1 associated DNA or protein complexes and have applications in studies related to kidney development.

Detailed description

The Osr1 gene encodes a transcription factor that is important in embryonic urogenital development. These transgenic mice express Osr1 with a Bio-FLAG tag on the 3’ terminus as well as a nuclear localized EGFP-BirA enzyme. The transgene expression pattern mimics the endogenous Osr1 gene expression pattern in embryos. Hemizygotes are viable and fertile. Homozygous viability/fertility has not been tested (July 2018).

Development

A transgenic construct containing Osr1 cDNA, BioTag-3XFLAG sequence, and an IRES-NLS-GFP-BirA cassette under the control of the Hsp68 (Hspa1a, heat shock protein 1A, mouse) minimal promoter, was injected into (C57BL/6 x DBA) F1 donor eggs. Founder line 1 was subsequently established. The mice were then backcrossed to C57BL/6J for many years. During backcrossing, the Y chromosome may not have been fixed to the C57BL/6J genetic background Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.
