Description

Cd36^flox mice possess loxP sites flanking exons 3-4 of the Cd36 gene. These floxed mice are useful when studying the role of CD36 in cellular processes including angiogenesis, inflammatory response and lipid metabolism.

Detailed description

Cd36^flox mice possess loxP sites flanking exons 3-4 of the CD36 molecule gene (Cd36). Cd36, also known as cluster of differentiation 36, encodes an integral membrane protein found on the surface of many cell types. It is a multifunctional immuno-metabolic receptor with many ligands that is involved in apoptotic cell uptake, signal transduction, cellular adhesion, angiogenesis, lipid metabolism, and immune function. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 3-4 deleted in the cre-expressing tissues.

When bred to Tie2-cre mice, expressing Cre recombinase in endothelial cells (ECs), EC-Cd36^-/- mice exhibit increased fasting plasma (FAs) and postprandial triglycerides, reduced uptake of radiolabeled long chain FAs into heart, skeletal muscle and brown adipose tissue, and improved glucose tolerance and insulin sensitivity.

 

Development

The Cd36^flox targeting vector was designed to insert a loxP site upstream of exon 3, and a frt-flanked neomycin selection cassette followed by a second loxP site downstream of exon 4 of the CD36 molecule (Cd36) gene. The construct was electroporated into (129xB6)F1-derived KV1 embryonic stem (ES) cells. Resulting ES cells were injected into C57BL/6 blastocysts and resulting Cd36^flox males were bred to ACTB-Flpe females on a C57BL/6 background to remove the neo cassette. Progeny were crossed to remove the Flpe-expressing transgene, and offspring were bred to C57BL/6 mice for at least six generations by the donating lab (see SNP note below). Upon arrival at The Jackson Laboratory Repository, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).

A 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. Two of the 43 markers throughout the genome were segregating with 129. Also, 1 of the 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest an incomplete backcross.
