Description

Sned^- mice carry a targeted knock-out of the mouse Sned1 gene in which exons 3 and 4 are deleted. Most homozygotes die within 2 days of being born. The rare null survivors present severe cranio-facial defects (shorter snouts, asymmetrical nasal cavities) and growth retardation.

Detailed description

Sned^- targeted knock-out mice have a deletion of exons 3 and 4 of the sushi, nidogen and EGF-like domains 1 gene (Sned1). Heterozygotes are viable, fertile, and do not demonstrate any overt phenotype. Most homozygotes die within 2 days of being born. The rare null survivors present severe cranio-facial defects (shorter snouts, asymmetrical nasal cavities) and growth retardation - phenocopying Sned1^LacZ-Neo mice (see Stock No. MMRRC 65813).

Development

This tm1d mutant strain is derived from a KOMP (Knockout Mouse Project) embryonic stem (ES) cell line (EPD0300_5_02 E02, subclone D2; Project ID: CSD34623). A vector introduced to C57BL/6N-derived JM8.N4 embryonic stem (ES) cells placed an FRT-En2 SA-IRES-lacZ-pA-loxP-hBactP-neo-pA-FRT-loxP cassette in intron 2, and a third loxP site in intron 4 of the Sned1 gene. Live mice were derived from the ES cells at the Massachusetts Institute of Technology. Resultant mice (see Stock No. MMRRC 65813) were crossed with B6.Cg-Tg(ACTFLPe)9205Dym/J animals to excise the FRT-flanked region, leaving exons 3 and 4 flanked by loxP sites (see Stock No. MMRRC 65814). Mice were then crossed with B6.C-Tg(CMV-cre)1Cgn/J mice to excise exons 3 and 4 to create widespread knock-out of the targeted gene. This strain was maintained on a genetic background incorporating C57BL/6N and C57BL/6J by the donating laboratory.

Molecular note: The L1L2_Bact_P cassette was inserted at position 93186212 of Chromosome 1 upstream of the critical exon(s) (Build GRCm39). The cassette is composed of an FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by a neomycin resistance gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of the targeted exon(s) at position 93187769. The critical exon(s) is/are thus flanked by loxP sites. A "conditional ready" (floxed) allele was created by flp recombinase expression in mice carrying this allele. Subsequent cre expression resulted in a knockout mouse.
