Sned1LacZ-Neo mice carry a knock-out first, conditional-ready, lacZ-tagged allele of the mouse Sned1 (sushi, nidogen and EGF-like domains 1) gene. This allele has an FRT-lacZ-loxP-Neo-FRT-loxP cassette in intron 2, and a third loxP site in intron 4. Expression of Sned1 is eliminated. Heterozygotes are viable, fertile, and do not demonstrate any overt phenotype. Most homozygotes die within 2 days of being born. The rare knock-out survivors present severe craniofacial defects (shorter snouts, asymmetrical nasal cavities) and growth retardation. Cre recombinase-mediated excision deletes the neomycin cassette and the floxed exons (3 and 4) and generates a lacZ-tagged allele. Flp recombinase-mediated excision of the FRT-flanked region (lacZ-neo cassette) creates a conditional allele in which exons 3 and 4 are flanked by loxP sites and expression of the targeted gene is restored, but there is no lacZ feature (see Stock No. <a href="https://www.jax.org/strain/032247">MMRRC 65814</a>).
