H1f0 (H1 histone family, member 0; also called H1.0) is expressed in terminally differentiated cells including neurons and encodes a protein that binds to linker DNA and compacts nucleosomes, causing chromatin structural changes. 

These Fl-H1.0 mice carry a FLAG-tagged mouse H1F0 transgene that can be conditionally expressed from a tetracycline-responsive promoter element (TRE, tetO). When the mice are crossed with mice expressing tTA (Tet-Off) or rtTA (Tet-On), resulting compound mutant pups will express the transgene in the absence (tTA) or presence (rtTA) of doxycycline in tissues of interest. 

When crossed with a Camk2a-tTA line (see Stock No. <a href="https://www.jax.org/strain/016198"">016198</a>), 1.5x over-expression of H1F0 protein is observed in forebrain excitatory cells. No phenotype is detected in the compound hemizygous mutant mice. Expression of H1F0 shows no measured effect on the expression of the Mecp2 gene. The independence of DNA binding by H1F0 and MECP2 in the forebrain has been further demonstrated through crosses involving MECP2-deficient (see Stock No. <a href="https://www.jax.org/strain/003890"">003890</a>) mice.

When crossed with a tTA- or rtTA-expressing strain, these Fl-H1.0 transgenic mice express FLAG-tagged mouse H1f0 (H1 histone family, member 0; also called H1.0) from the tetO (TRE) tetracycline-responsive promoter element. This strain is useful in studies of chromatin structural changes.
