Description

DR3^FL mice possess loxP sites flanking exons 1-5 of the tumor necrosis factor receptor superfamily, member 25 (Tnfrsf25) gene. These floxed mice are useful for tissue specific deletion of DR3 when studying lymphocyte stimulation and autoimmune inflammation.

Detailed description

DR3^FL mice possess loxP sites flanking exons 1-5 of the tumor necrosis factor receptor superfamily, member 25 (Tnfrsf25) gene. DR3 is expressed primarily on lymphocytes and is a receptor for TNF superfamily member, TL1A. TL1A is a cytokine implemented in the onset and development of autoimmune and autoinflammatory diseases such as inflammatory bowel disease and rheumatoid arthritis. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 1-5 deleted in the cre-expressing tissues.

Development

The DR3^FL targeting vector was designed to insert loxP sites flanking exons 1-5 of the tumor necrosis factor receptor superfamily, member 25 (Tnfrsf25) gene, and a frt-flanked neomycin selection cassette was also inserted. The construct was electroporated into C57BL/6NTac-derived ART B6 3.6 embryonic stem (ES) cells and resulting ES cells were injected into blastocysts. The resulting DR3^FL mice were bred to C57BL/6NTac mice and progeny were crossed to Flp recombinase expressing mice on a C57BL/6 background to remove the neo cassette. Offspring were crossed to remove the Flp-expressing transgene, and offspring were bred to C57BL/6NTac mice. Upon arrival at The Jackson Laboratory Repository, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).
