Description

Exon 1 of the mouse Trip11 gene is flanked by loxP sites in this Trip11^cko conditional mutant strain. Cre-mediated excision of the floxed region results in a knock-out allele, useful in studies of achondrogenesis type 1A (ACG1A).

Detailed description

Trip11 (thyroid hormone receptor interactor 11) encodes ubiquitously-expressed membrane trafficking component GMAP-210. Homozygous mutations in the protein cause achondrogenesis type 1A (ACG1A), a perinatal lethal condition which primarily affects cartilage development.

Exon 1 of the mouse Trip11 gene is flanked by loxP sites in this Trip11^cko conditional mutant strain. Cre-mediated excision of the floxed region results in a knock-out allele.

Comparable levels of TRIP11 protein are expressed by the Trip11^cko floxed allele and Trip11 wildtype allele, as determined by western blot analysis of mouse embryonic fibroblasts. Animals homozygous for the conditional allele are indistinguishable from wildtype and heterozygous conditional mutant mice.

Crosses with germline-specific EIIa-cre mice (see Stock No. 003724) produce homozygous embryos (E17.5) that have the same skeletal dysplasia previously observed in mice with other loss-of-function alleles. They feature a short trunk, short limbs, and a domed skull with protruding tongue. A smaller rib cage, decreased mineralization of the calvarium, absent mineralization of the sternum, and absent mineralization of the vertebral bodies are also seen.

Osteoblast-specific excision of the floxed region via crosses with BGLAP-cre mice (see Stock No. 019509) does not result in skeletal dysplasia.

Development

A loxP site was inserted 671 bp upstream of exon 1 (which contains the ATG) and an FRT site-flanked neomycin selection cassette, immediately followed by a loxP site, was placed 1506 bp downstream of exon 1. The mutation was created in 129Sv-derived embryonic stem (ES) cells. Resultant mice were crossed with FLPeR mice (see Stock No. 009086) to excise the neomycin cassette, leaving exon 1 flanked by loxP sites. This strain was maintained on a mixed C57BL/6J and 129/Sv genetic background by the donating laboratory.
