Description

POU6f1^{floxed Exon2-11} mice possess loxP sites flanking exons 2-11 of the POU domain, class 6, transcription factor 1 gene. These floxed mice are useful for tissue specific deletion of Pou6f1 when studying neuronal plasticity in the adult brain.

Detailed description

POU6f1^{floxed Exon2-11} mice possess loxP sites flanking exons 2-11 of the POU domain, class 6, transcription factor 1 (Pou6f1) gene. POU6f1 is a brain specific transcription factor involved in dendrite specification and patterning, as well as synapse development and function in adult-born neurons. The floxed region in these mice includes the DNA binding domain. Mice that are homozygous for this floxed allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 2-11 deleted in the cre-expressing tissues.

When bred to mice expressing Cre recombinase in corticotropin-releasing hormone (CRH) receptor 1 (CRHR1)-expressing neurons, resulting mice exhibit reduced dendritic complexity and decreased synaptic connectivity in olfactory granule cells.

Of note, a CRISPR/cas9 endonuclease mediated mutant containing a 3xHA tag upstream of the Pou6f1 stop codon is available as Stock No. 031997.

Development

The POU6f1^{floxed Exon2-11} targeting vector was designed to insert loxP sites flanking exons 2-11 of the POU domain, class 6, transcription factor 1 (Pou6f1) gene, and an frt-flanked neomycin selection cassette was inserted downstream of exon 11. The construct was electroporated into 129S7/SvEvBrd-Hprt^b-m2-derived AB2.2 embryonic stem (ES) cells. Resulting ES cells were injected into C57BL/6 blastocysts and resulting POU6f1 cKO males were bred to C57BL/6 females. Subsequently, mice were bred to B6N.129S4-Gt(ROSA)26Sor^tm1(FLP1)Dym/J mice (Stock No. 016226) to remove the neo cassette. Progeny were crossed to remove the FLPe-expressing transgene, and offspring were maintained on a mixed genetic background. Upon arrival at The Jackson Laboratory Repository, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

Name: targeted mutation 1.1, Benjamin Arenkiel
MGI accession ID: MGI:6153800
Generation method: Targeted
Attributes: Conditional ready (e.g. floxed)
Synonyms: POU6f1 cKO
Marker symbol: Pou6f1
Marker name: POU domain, class 6, transcription factor 1
Marker synonyms: 2310038G18Rik; Brn5; cns-1; Emb; MPOU; TCFB1
Chromosome: 15
Strain of origin: 129S7/SvEvBrd-Hprt1^b-m2
Molecular note: The targeting vector contains a loxP site downstream of exon 1, and an FRT-flanked neomycin cassette followed by a loxP site downstream of exon 11. Flp-mediated recombination removed the FRT-flanked neo cassette.
