Description

HbE transgenic mice express mutant human hemoglobin E (beta26 Glu to Lys), exhibit mild hypochromic red blood cells and target cells, and increased hemoglobin auto-oxidation. These mice may be suitable for use in studies related to thalassemia, sickle cell anemia and hemoglobinopathy.

Detailed description

These HbE transgenic mice express human hemoglobin E, which has a β chain point mutation, E26K, resulting in a glutamic acid to lysine change at position 26, and the human α chain. Hemoglobin E (beta26 Glu to Lys) is the most common abnormal hemoglobin variant in the world. Transgenic mice exhibit mild hypochromic red blood cells and target cells. Hemoglobin auto-oxidation is higher in transgenic mice compared to noncarrier controls. Mice hemizygous for the transgene are viable and fertile. Homozygous viability/fertility has not been tested (February 2018). Genotyping assays developed by The Jackson Laboratory were validated by HPLC analysis.

Development

A transgenic insert containing the human alpha globin 2 (HBA2) gene from the locus control region (LCR) of the human beta globin gene and a second transgenic insert containing a mutant human beta globin (HBB) gene with the E26K point mutation (position 26 glutamic acid to lysine) were co-injected into fertilized FVB/N mouse eggs. Founder line 53 was subsequently established.

The mice were backcrossed to C57BL/6J for an unknown number of generations. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
