Description

Exon 4 of the mouse Tenm3 gene is flanked by loxP sites in this Ten3^flox conditional mutant strain. Cre-mediated excision of the floxed region results in a knock-out allele.

Detailed description

Teneurins are evolutionarily conserved transmembrane proteins that instruct synaptic partner matching in Drosophila and are required for vertebrate visual system development. Mouse teneurin-3 (Tenm3) is expressed in multiple topographically interconnected areas of the hippocampal region, including proximal CA1, distal subiculum, and medial entorhinal cortex. Viral-genetic analyses reveal that teneurin-3 is required in both CA1 and subicular neurons for the precise targeting of proximal CA1 axons to distal subiculum.

In this Ten3^flox CRISPR/Cas9-derived conditional mutant line, exon 4 of the mouse Tenm3 gene is flanked by loxP sites. Cre-mediated excision of the floxed region creates a frame shift and results in a knock-out allele (as demonstrated through crosses with Ten3-cre (see Stock No. 031704) or Emx1-cre mice, or via cre-expressing lentivirus injections).

Development

A reversed loxP site was introduced to intron 3 and an FRT-flanked PGK-neomycin resistance cassette and reversed loxP site were placed in intron 4 via CRISPR/Cas9 methodology using C57BL/6 x 129S6 F1 embryos. The neomycin cassette was removed by FlpO-mediated recombination via crosses with mice expressing Tg(CAG-GFP,Pgk1-flpo)1Luo on a CD1 genetic background. This strain was maintained on a mixed CD-1 (predominantly), C57BL/6, and 129 genetic background by the donating laboratory.
