The targeted Mtor gene encodes a serine/threonine protein kinase that is a major regulator of protein translation and synthesis and is involved in stress response, as well as cell growth and survival. Deregulation of the mTOR signaling pathway occurs in many types of cancer.
This CRISPR/Cas9-generated mutant of the Mtor gene carries a deletion in the Mtor 3' UTR sequence that confers axonal localization of the transcript. Expression of the deleted Mtor 3' UTR was confirmed by 3' RACE analysis of dorsal root ganglia from homozygotes. FISH analysis of sciatic nerve sections detects a significant reduction of Mtor mRNA. Isolated neurons from homozygotes exhibit significantly decreased mTOR protein levels in growth cones and axonal tips. Sciatic nerve sections show attenuation of mTOR protein levels upon injury as well as reduction in local protein synthesis and neuron survival in null mice. Homozygotes are viable and fertile.

mTOR 3'UTR null knock-out mice carry a CRISPR/Cas9-generated deletion in the Mtor 3' UTR sequence that confers axonal localization of the Mtor transcript. These mice are useful in studies of mTOR transcript localization.

Typically mice are recovered in 12-16 weeks. Contact Customer Service to place an order or for more information.
