Description

These Rosa^Lef1GFP mice are a useful for the overexpression and labeling of Lef1 in Cre Recombinase expressing cell lines.

Detailed description

These Rosa^Lef1GFP mice contain from 5' to 3', a CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid promoter (from pCAGGS), a floxed-STOP cassette, murine lymphoid enhancer binding factor 1 (Lef1) cDNA, T2- Green Fluorescent Protein (T2-mGFP), and a frt-flanked neomycin resistance (neo) cassette. This targeting vector was inserted into the Gt(ROSA)26Sor locus. The transcription factor encoded by the Lef1 gene is involved in T cell development and the Wnt signaling pathway, and has been associated with tumorigenesis. The donating investigator has not attempted to make the colony homozygous.

In the absence of Cre, gene expression is prevented by the STOP cassette. After removal of the loxP-flanked STOP cassette via cre-mediated recombination, Lef1 and T2-mGFP are expressed, allowing for labeling cells/tissues where Cre Recombinase is expressed.

For example, when expressed in Tcf4-expressing cells of the gut, mice die shortly after birth due to loss of cell homeostasis in the colon.

Development

The Rosa^Lef1GFP targeting vector was designed with (from 5' to 3') a ubiquitously expressed CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid promoter (from pCAGGS), loxP-flanked stop cassette, murine lymphoid enhancer binding factor 1 (Lef1) cDNA, a membrane bound T2-Green Fluorescent Protein (T2-mGFP), and a neomycin resistance (neo) cassette. This targeting vector was inserted into the Gt(ROSA)26Sor locus. The construct was electroporated into (129S6/SvEvTac x C57BL/6NCrl)F1-derived G456 embryonic stem (ES) cells and correctly targeted ES cells were injected into blastocysts. The resulting chimeric animals were crossed to C57BL/6J mice for at least 10 generations. Upon arrival at The Jackson Laboratory, the mice were crossed to C57BL/6J mice (Stock No. 000664) at least once to establish the colony.
