Description

Tcf4^Lox mice possess loxP sites flanking exon 1 of the Tcf7l2 gene, making them useful when making tissue-specific Tcf4 knock-outs.

Detailed description

Tcf4^Lox mice have loxP sites flanking exon 1 of the transcription factor 7 like 2, T cell specific, HMG box gene (Tcf7l2). Tcf7l2 is a transcription factor that plays a role in the Wnt signaling pathway and is involved in the maintenance of the epithelial stem-cell compartment of the small intestine. Mutations in this gene have been associated with colon carcinogenesis. Expression is also evident in connective tissue fibroblasts. Mice that are homozygous for this allele are viable and fertile. When Tcf4^Lox are bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 1 deleted in the cre-expressing tissues.

Development

The Tcf4^Lox targeting vector was designed to insert loxP sites flanking exon 1 of the transcription factor 7 like 2, T cell specific, HMG box gene (Tcf7l2), followed by a frt-flanked neomycin resistance (neo) cassette. The construct was introduced into (129S6/SvEvTac x C57BL/6NCrl)F1-derived G456 embryonic stem (ES) cells, and correctly targeted ES cells were injected into blastocysts. Chimeric males were bred to C57BL/6 mice and offspring were bred to ACTB:FLPe mice (Stock No. 005703) to remove the neo cassette. Resulting Tcf4^Lox mice were bred to C57BL/6J mice (Stock No. 000664) for 15 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.

Name: targeted mutation 2.1, Mario R Capecchi
MGI accession ID: MGI:4946910
Generation method: Targeted
Attributes: Conditional ready (e.g. floxed); No functional change
Synonyms: Tcf4^Lox; Tcf4^loxEx1Lox
Marker symbol: Tcf7l2
Marker name: transcription factor 7 like 2, T cell specific, HMG box
Marker synonyms: mTcf-4B; mTcf-4E; Tcf4; TCF-4; TCF4B; TCF4E
Chromosome: 19
Strain of origin: (129S6/SvEvTac x C57BL/6NCrl)F1
Molecular note: Exon 1 was replaced with a floxed exon 1 and an FRT-flanked neo cassette. Flp-mediated recombination removed the neo cassette.
General note: ES cell line = G456
