Description

The BAC C transgenic strain expresses lacZ reporter from Bmp5 regulatory elements and is relevant to studies of bone development and gene regulation.

Detailed description

This strain was built to investigate potential regulatory sequences surrounding the mouse Bmp5 (bone morphogenetic protein 5) gene, which signals the formation of particular anatomical features in the skeleton during development.

During embryonic development, BAC C, encoding regulatory elements that control expression of mouse Bmp5, drives expression of a lacZ reporter in the medial perichondrium of developing ribs. BAC C also controls expression of lacZ in many additional developing tissues including the fingertips, the cartilages of the inner ear, the tip of the nasal region, the manubrium, lung mesenchyme, thyroid cartilage and the tip of the genital tubercle. Retinal expression controlled by the Hsp68 (Hspa1b) promoter is also visible. In adults, BAC C drives lacZ expression following injury to multiple tissues including bone, skin and lung.

Development

Linearized BAC C clone (RP23-476E23), encoding regulatory elements that control expression of mouse Bmp5 (but no Bmp5 coding sequences) as well as all but the last two exons of the Hmgcll1 gene, was isolated from the RPCI-23 female (C57BL/6J) mouse BAC library and co-injected into FVB/N embryos with a minimal Hsp68 (Hspa1b)-lacZ reporter. Resultant mice were crossed to FVB/NJ for more than 6 generations by the donating laboratory. The insertion site and copy number of the transgene is not known.

Expressed genes: lacZ,beta-galactosidase,E. coli
Site of expression: The lacZ reporter is expressed in the medial perichondrium of developing ribs.
Molecular note: The linearized BAC clone (RP23-476E23) encodes regulatory elements of mouse Bmp5 (but no coding sequences) and the last two exons of the Hmgcll1 gene; it was co-injected with a minimal hsp68-lacZ reporter. The clone was isolated from the RPCI-23 female (C57BL/6J) mouse BAC library and contains a sequence that controls expression of the medial perichondrium of embryonic ribs. Robust expression is observed at E14.5 dpc.
