Description

The BAC A transgenic strain expresses lacZ reporter from a genomic fragment incorporating the complete coding sequences of the mouse Bmp5 gene. It is useful in studies of bone development and gene regulation.

Detailed description

This strain was built to investigate potential regulatory sequences surrounding the mouse Bmp5 (bone morphogenetic protein 5) gene, which signals the formation of particular anatomical features in the skeleton during development.

During embryonic development, BAC A, encoding the complete coding sequences of mouse Bmp5 (and no other genes), drives expression of a lacZ reporter in the lateral perichondrium of developing ribs, in the perichondrium of developing forelimb and hindlimb bones, and in the nasal sinus. Retinal expression of lacZ, controlled by an Hsp68 (Hspa1b) minimal promoter, is also visible.

Development

Linearized BAC A clone (RP23-377M6), encoding the complete coding sequences of mouse Bmp5 (and no other genes), was isolated from the RPCI-23 female (C57BL/6J) mouse BAC library and co-injected into FVB/N embryos with a minimal Hsp68 (Hspa1b)-lacZ reporter. Resultant mice were crossed to FVB/NJ for more than 6 generations by the donating laboratory. The insertion site and copy number of the transgene is not known.
