Exon 2 of the mouse Epn1 (epsin 1) gene is flanked by loxP sites in this Epn1fl line. Cre-mediated excision of the floxed region results in a knock-out allele which has been useful in studies of vascular endothelium, tumor growth, cell division, and endocytosis.

Exon 2 of the mouse Epn1 (epsin 1) gene is flanked by loxP sites in this Epn1fl line. Homozygous floxed mice are viable and fertile, without any overt phenotype. Cre-mediated excision of the floxed region results in a knock-out allele. Mice carrying a widespread knock-out of the gene are reported to show no significant phenotype.
Widespread knock-out of Epn1 combined with a complete knock-out of Epn2 (see Stock No. 031166) results in embryonic lethality at approximately E9.5.
Mice carrying both a knock-out of Epn2 and a vascular endothelial knock-out of Epn1, achieved via crosses with a tamoxifen-inducible Cdh5 (VEcadherin)-cre strain, demonstrate decreased tumor growth. A 50% reduced incidence at 1 week is observed in animals subcutaneously implanted mouse Lewis lung carcinoma (LLC) or melanoma cells. Reduced growth of gliomas, colorectal tumors, and prostate tumors have also been shown in compound mutant mice.
Triple knock-out mice derived from this Epn1 floxed line, Epn2 knock-out mice (see Stock No. 031166), and Epn3 knock-out mice (see Stock No. 014108) are also embryonic lethal. Their cells display a dramatic cell division defect and a robust impairment in clathrin-mediated endocytosis.
A loxP site was placed in intron 1 and an FRT-neomycin-FRT-loxP site was introduced to intron 2 via homologous recombination in an unknown embryonic stem (ES) cell line. Though crossed with a Flp-expressing strain by the donating laboratory, genotyping assays at The Jackson Laboratory suggest that the neomycin cassette was retained in the imported mice. The donating laboratory indicates a mixed C57BL/6 and 129 genetic background.
