Description

These p75^NTR-FX, mutant mice possess loxP sites flanking exons 4 through 6 of the Ngfr gene. This strain may be useful for generating conditional mutations in applications related to neuronal electrophysiology, neuronal progenitor cell cycle and neural cell survival.

Detailed description

The targeted Ngfr gene, also known as p75^NTR, encodes a transmembrane nerve growth factor receptor that is involved in cell proliferation and survival, synaptic plasticity, neurite outgrowth, myelination, glucose and lipd metabolism, and circadian rhythm. These mice possess loxP sites flanking exons 4 through 6 of the targeted Ngfr gene. Exons 4 through 6 encode the transmembrane and cytoplasmic domains of the protein. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exons 4 through 6 deleted in the cre-expressing tissues. Removal of the floxed sequence creates a null allele.

When bred to a strain with Cre recombinase expression in the germline (see Stock No. 003724 for example), this mutant mouse strain may be useful in studies of neural development.

Development

A targeting vector containing loxP site flanked PGK-Neo selection cassette was utilized in the construction of this mutant. This selection cassette was inserted downstream of exon 6 of the targeted gene, and another loxP site was inserted upstream of exon 4. This construct was electroporated into 129S2/SvPas derived TVB2 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts. The resulting chimeric animals were tested for germline transmission. Heterozygotes were intercrossed to generate homozygotes. Female homozygous mice were then crossed to mice expressing Cre recombinase under the control of the adenovirus EIIa promoter (Stock No. 003724) to excise the selection cassette. Mice that no longer carried the selection cassette but did retain the loxP-flanked exons 4 through 6 were bred to remove the Cre recombinase transgene. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
