α5^fl mice possess loxP sites flanking exons 4-5 of the gamma-aminobutyric acid (GABA) A receptor, subunit alpha 5 (Gabra5) gene, making them useful for tissue specific deletion of α5-subunit containing GABA_A receptors (α5-GABA_ARs). These floxed mice may have applications related to neurotransmission.

Detailed description

α5^fl mice possess loxP sites flanking exons 4-5 of the gamma-aminobutyric acid (GABA) type A receptor, subunit alpha 5 (Gabra5) gene. Gabra5 encodes the α5 subunit of the GABA_A receptor family, which are ligand-gated ion channels. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 4-5 deleted in the cre-expressing tissues.

Development

The α5^fl targeting vector was designed to insert a single loxP site downstream of exon 5, and a loxP-NEO-TK-loxP cassette downstream of exon 5 of the gamma-aminobutyric acid (GABA) A receptor, subunit alpha 5 (Gabra5) gene. The construct was electroporated into C57BL/6N-derived embryonic stem (ES) cells. Correctly targeted ES cells transiently transfected with a cre-expressing plasmid to remove the neo cassette. Resulting ES cells were injected into blastocysts and resulting α5^fl mice were bred to C57BL/6J mice for at least 10 generations. Frozen sperm was sent to The Jackson Laboratory Repository. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
