Detailed description

Tnni3k KO mice lack exons 1-2 of the TNNI3 interacting kinase (Tnni3k) gene, abolishing gene expression. Tnni3k is a cardiomyocyte-specific enzyme in the MAP kinase kinase kinase (MAPKKK) family. It is thought to play a role in cardiac physiology where it promotes ischemia/reperfusion injury, oxidative stress, and myocyte death. Mice that are homozygous for this allele are viable and fertile. They have elevated mononuclear diploid cardiomyocyte content and increased cardiomyocyte proliferation after ischemic injury. Mononuclear diploid cardiomyocytes are a population of proliferative cardiomyocytes in the adult heart.

Of note, other inbred lines contain a natural Tnni3k null allele, including DBA/2J, C3H/HeJ and BALB/cByJ.

Development

A targeting vector was designed to insert loxP sites flanking exons 1-2 of the TNNI3 interacting kinase (Tnni3k) gene, and a neomycin selection cassette was inserted. The construct was electroporated into embryonic stem (ES) cells. Resulting ES cells were injected into blastocysts and resulting Tnni3k^fl mice were bred to C57BL/6J mice. Some mice were subsequently bred to Sox2-cre mice (see Stock No. 008454) to remove the neo cassette and exons 1-2. Progeny were crossed to remove the Cre-expressing transgene, and offspring were bred to C57BL/6J mice for at least 2 more generations. Upon arrival at The Jackson Laboratory Repository, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
