Stat3 (signal transducer and activator of transcription 3) encodes a transcription activator induced by cytokines and growth factors; it has roles in inflammation, development and cancer. The TRE-Stat3.1348 (Stat3 shRNA) allele is comprised of a TRE;tetO promoter controlling expression of a Stat3 short hairpin RNA and a GFP reporter. This doxycycline-mediated, reversible allele allows the silencing of Stat3 by RNA interference. When combined with a reverse transactivator under the control of the widespread CAG promoter and administration of doxycycline, activation of Stat3 shRNA reduces Stat3 expression by 80% in the gastrointestinal tract as well as pancreas, heart, lung and liver, but not spleen. GFP expression is observed in the epithelial cells of the stomach and inversely correlates with the presence of STAT3 protein. In a mouse model of gastric cancer, induction of Stat3 shRNA reduces tumor burden in the stomach. In the embryo, induction of Stat3 shRNA results in partial embryonic lethality. This TRE-Stat3.1348 strain is a valuable tool to study the use of STAT3 therapeutic inhibition in multiple diseases and especially in gastric cancers.
