Description

These BAC transgenic mice express cre recombinase from the mouse Colgalt2 gene. Recombination has been observed in striatal-projecting pyramidal neurons of the isocortex and in pyramidal neurons in the piriform, subicular and lateral entorhinal cortices. This mouse line may be useful to investigators who want to study neuronal projections from the cerebral cortex to the striatum.

Detailed description

These BAC transgenic mice express cre recombinase from the mouse Colgalt2 gene. Recombination has been observed in striatal-projecting pyramidal neurons of the isocortex and in pyramidal neurons in the piriform, subicular and lateral entorhinal cortices. Expression is found in the axons of vestibulocochlear and optic nerves. The endogenous Colgalt2 gene is not perturbed by the mutation.

Used in conjunction with an injected cre-dependent adeno-associated virus (AAV) expressing the ribosomal L10-GFP fusion protein, cre-expressing cells can be effectively labeled in layer 5 neurons of the medial prefrontal cortex (mPFC).

Development

Mouse BAC RP23-160M1 (including partial sequences of Rgl1, and complete sequences of both Cogalt2 and Tsen15) was modified using a two-plasmid/one-recombination protocol to incorporate cre recombinase in a region just upstream of the Colgalt2 ATG start codon. The modified BAC was microinjected into the pronucleus of an FVB/N zygote and randomly integrated into the mouse genome. The endogenous Colgalt2 gene was not perturbed. Resultant mice were bred to CD1, then to C57BL/6J for 2 generations by the donating laboratory. This is founder line number NF111.
