Tet3 (tet methylcytosine dioxygenase 3) produces oxidized methylcytosines, intermediates in DNA methylation. Knock-outs of the gene, on either a C57BL/6 or 129Sv genetic background, cause perinatal lethality. These Tet3fl mice carry a conditional mutant allele in which exon 2 of the mouse Tet3 gene is flanked by loxP sites. Cre recombinase-mediated excision of the floxed region results in a knock-out allele. The knock-out allele produces no detectable protein as judged by immunocytochemistry of Tet3-deficient 2-cell embryos using an antibody (Gene Tex 121453) which recognises an epitope in the C-terminus, outside the deleted area; RNA for the undeleted exons can be present.
