Description

Il22ra1^fl mice possess loxP sites flanking exon 3 of Il22ra1 making them useful for tissue specific IL-22ra1 deficiency. These floxed mice are useful when studying host defense against extracellular pathogens.

Detailed description

Il22ra1^fl mice possess loxP sites flanking exon 3 of the interleukin 22 receptor, alpha 1 (Il22ra1) gene. IL-22RA1 is a subunit of the IL20, IL22 and IL24 receptors. IL-22RA1 is expressed in epithelial cells and hepatocytes where it plays a critical role in host defense against extracellular pathogens, particularly in the intestine. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 3 deleted in the cre-expressing tissues.

When bred to B6.Cg-Tg(Alb-cre)21Mgn/J mice (Stock No. 003574) expressing Cre Recombinase in the liver, resulting IL-22RA1 deficient mice have higher bacterial burdens in the lung after intrapulmonary pneumococcal infection.

 

Development

The Il22ra1^fl targeting vector was designed to insert a loxP site upstream of exon 3 and a frt-flanked neomycin resistance (neo) cassette, followed by a second loxP site, was inserted downstream of exon 3 of the interleukin 22 receptor, alpha 1 (Il22ra1) gene. The construct was electroporated into B6(Cg)-derived Bruce4 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric mice were bred with C57BL6/J mice (Stock No. 000664). Offspring were bred to Flp Recombinase expressing mice to delete the neo cassette. Progeny were bred with C57BL/6J mice to remove the Flp-expressing transgene. Resulting Il22ra1^fl/fl mice were bred with C57BL/6J mice for at least 5 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.
