Description

These floxed mutant mice possess loxP sites flanking exon 2 of the Rhoq gene. This strain may be useful for generating conditional mutations in applications related to actin cytoskeleton reorganization, the immune synapse, and the insulin pathway.

Detailed description

The targeted Rhoq gene encodes protein that is a member of the Rho subfamily of the RAS superfamily of small GTP-binding proteins. It is involved in reorganization of the actin cytoskeleton and regulates cell shape, polarity, attachment, and motility. RHOQ is importation for formation of filopodia, axonal growth and plays a role in the immune synapse as well as the insulin pathway.

These mice possess loxP sites on either side of exon 2 of the targeted gene. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 2 deleted in the cre-expressing tissues. Removal of the floxed sequence creates a null allele.

When bred to a strain with widespread Cre recombinase expression (see Stock No. 020811 for example), this mutant mouse strain may be useful in studies of B cell development and B cell receptor activation.

Development

A targeting vector containing a FRT site flanked neomycin (neo) selection cassette was utilized in the construction of this mutant. This selection cassette and a 3’ loxP site was inserted downstream of exon 2 of the targeted gene, and another loxP site was inserted upstream of exon 2. This construct was electroporated into unspecified C57BL/6J derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into BALB/c blastocysts.

The resulting chimeric animals were tested for germline transmission. Mutant mice were crossed to a FLP recombinase expressing strain on the C57BL/6 background to remove the FRT site flanked neo cassette. Mice that no longer contained the neo selection cassette and retained floxed exon 2 were backcrossed to C57BL/6J for 3 or 4 generations. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
