Detailed description

GPER1 (G protein-coupled estrogen receptor 1 or GPR30) is a membrane protein that localizes to the endoplasmic reticulum and binds estrogen thereby mediating intracellular calcium mobilization. These knock-out mice contain a neomycin cassette used to replace exon 3. Mice homozygous for the mutation are viable and fertile. Of note, The Jackson Laboratory live colony shows homozygous matings may not be as productive as heterozygous matings [2019].
Homozygous female mice implanted with 17β-estradiol (E2) exhibit a reduction in thymic atrophy as compared to wild-type females. Specifically, E2-induced apoptosis in TCRβ double positive (DP) thymocytes, but not double negative (DN) thymocytes, is reduced. This strain may be useful for studying the estrogen-induced signaling pathways in thymic development.

Development

A targeting vector containing a neomycin resistance (neo) cassette was used to replace exon 3. The construct was electroporated into 129S6/SvEvTac-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric mice were bred to C57BL/6J mice for at least 5 generations to establish a colony. Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6J inbred mice for at least one generation.

A 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. One of the 43 markers, on Chromosome 12, was segregating.
