When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 1 deleted in the cre-expressing tissues.
Progeny produced by combining Srebf2fl mice and mice carrying the liver specific albumin-cre transgene (Tg(Alb-cre)21Mgn) exhibit decreased plasma and liver cholesterol concentrations as well as plasma and liver tryiglyceride levels.
The targeting vector contains a loxP site inserted in the promoter region 660 bp upstream of exon 1, and a loxP site followed by a FRT-flanked neomycin cassette inserted into intron 1. The construct was electroporated into 129S6/SvEv derived SM-1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting chimeric animals crossed to C57BL/6J and offspring were crossed to C57BL/6J for an unknown number of generations. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
