Detailed description

Pecam1^fl/fl mice possess loxP sites flanking exons 1-2 of the platelet/endothelial cell adhesion molecule 1 (Pecam1) gene. PECAM-1 is a 130-kDa member of the immunoglobulin gene superfamily (Ig) that is present on most cells of the hematopoietic lineage including platelets, monocytes, neutrophils and certain lymphocyte subsets, and is highly expressed at the junctions of confluent endothelial cell monolayers. PECAM-1 serves as an inhibitory receptor in circulating platelets and leukocytes, and functions in leukocyte transendothelial migration, and as an adhesive stress-response protein to both maintain endothelial cell junctional integrity and speed restoration of the vascular permeability barrier following inflammatory or thrombotic challenge. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, the resulting offspring will have exons 1-2 deleted in the cre-expressing tissues.

Development

A PECAM-1^fl/fl targeting vector was designed to insert a loxP site upstream of exon 1 and an FRT-flanked neomycin resistance (neo) cassette, followed by a second loxP site, downstream of exon 2 of the platelet/endothelial cell adhesion molecule 1 (Pecam1) gene. The construct was electroporated into C57BL/6-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts and the resulting chimeric mice were bred with B6.Cg-Tg(ACTFLPe)9205Dym/J transgenic mice (Stock No. 005703) to delete the neo cassette. Progeny were crossed to remove the Flp-expressing transgene. The resulting PECAM-1^fl/fl mice were bred to and maintained on a C57BL/6J congenic background. Upon arrival at The Jackson Laboratory Repository, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
