Description

These Ca_vα₂δ₁ floxed mutant mice possess loxP sites flanking exon 6 of the Cacna2d1 gene. This strain may be useful for generating conditional mutations in applications related to nociception and peripheral nerve injury.

Detailed description

The targeted Cacna2d1 gene encodes the alpha-2/delta-1 subunit of the voltage-dependent calcium channel complex found in muscle, brain and endocrine tissues. These mice possess loxP sites on either side of exon 6 of the targeted Cacna2d1 gene. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 6 deleted in the cre-expressing tissues. Removal of the floxed sequence creates a null allele.

When bred to a strain with Cre recombinase expression in Advillin-positive sensory neurons (see Stock No. 010745 for example), this mutant mouse strain may be useful in studies of nociception.

Development

A targeting vector containing a FRT-site flanked neo selection cassette was utilized in the construction of this mutant. This selection cassette and a loxP site was inserted downstream of exon 6 of the targeted gene, and another loxP site was inserted upstream of exon 6. This construct was electroporated into (129X1/SvJ x 129S1/Sv)F1- Kitl⁺-derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts.

The resulting chimeric animals were crossed to transgenic mice (on the C57BL/6 genetic background) expressing FLP recombinase. Mice that retained the loxP site flanked exon 6 were then bred to C57BL/6J mice to remove the FLP allele/ transgene. The mice were backcrossed to C57BL/6J for 20 generations.

Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
