Description

Tie2-CreER^T2 mice express a tamoxifen-inducible Cre recombinase gene under direction of the TEK receptor tyrosine kinase (Tek) promoter on a BAC transgene. They are useful for applications requiring tamoxifen-induced deletion of floxed sequences in endothelial cells.

Detailed description

Tie2-CreER^T2 mice express a tamoxifen-inducible codon improved Cre recombinase gene under direction of the TEK receptor tyrosine kinase (Tek) promoter on a BAC transgene. These mice express Cre-ER^T2 protein in endothelial cells. Restricted to the cytoplasm, Cre-ER^T2 can only gain access to the nuclear compartment after exposure to tamoxifen. When these mice are bred with mice containing loxP-flanked sequence, tamoxifen-inducible, cre-mediated recombination will result in deletion of the floxed sequences in the cre-expressing endothelial cells of the offspring. Mice hemizygous for the Tie2-CreER^T2 transgene are viable and fertile. The donating lab has not attempted to make this strain homozygous.

Development

A tamoxifen-inducible codon improved Cre recombinase gene was inserted into the ATG start site of the murine TEK receptor tyrosine kinase (Tek) gene located on a bacterial artificial chromosome (BAC). The construct was linearized and microinjected into fertilized FVB/N oocytes. Tie2-CreER^T2 founder mice were bred to C57BL/6J mice for at least 7 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).

Cre expression was detected in endothelial cells of numerous organs after treatment with tamoxifen.
