Description

These floxed mutant mice possess loxP sites flanking exons 1 and 2 of the Col15a1 gene. This strain may be useful for generating conditional mutations in applications related to atherosclerosis, basement membranes and fibrillary collagen fiber myopathy.

Detailed description

Collagen type 15 alpha chain, encoded by the Col15a1 gene, is a chondroitin sulfate modified glycoprotein found in the basement membranes zones of the extracellular matrix. These mice possess loxP sites on either side of exons 1 and 2 of the targeted gene. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exons 1 and 2 deleted in the cre-expressing tissues. Removal of the floxed sequence creates a null allele.

When bred to a strain with Cre recombinase expression in smooth muscle cells (see Stock No. 019079 for example) and Apoe knock-out mice (see Stock No. 002052 for example), this mutant mouse strain may be useful in studies of advanced atherosclerotic lesion formation.

Development

A targeting vector containing a FRT-site flanked PGK-neo-bGHpA cassette was utilized in the construction of this mutant. This selection cassette and a loxP site was inserted downstream of exon 2 of the targeted gene, and another loxP site was inserted upstream of exon 1. This construct was electroporated into C57BL/6N derived C57BL/6N-PRX-B6N #1 (Primogenix) embryonic stem (ES) cells. Correctly targeted ES cells were injected into albino C57BL/6J blastocysts. The resulting chimeric animals were crossed to congenic C57BL/6J FLPe deleter mice (Stock No. 009086) to excise the FRT-site flanked selection cassette. Mice that retained the loxP site flanked exons 1 and 2 were then bred to C57BL/6 mice to remove the FLPe recombinase allele. Heterozygotes were crossed to generate homozygotes. The mice were crossed to mice on a B6.129P2 genetic background and to mice on a B6.FVB genetic background. The mice were crossed to C57BL/6J once. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
