In this bacTRAP strain, the Abcb1a (ATP-binding cassette, sub-family B (MDR/TAP), member 1A) promoter drives the expression of EGFP-tagged ribosomal protein L10a (EGFPL10a) in endothelial cells throughout the central nervous system and the rest of the body. Expression of endogenous Abcb1a is expected to be knocked out. EGFPL10a expression allows for the isolation of tagged ribosomes by the translating ribosome affinity purification (TRAP) technique which can be used to access cell type-specific, polysome-bound transcripts in vivo from whole tissue homogenates. Native EGFP fluorescence or immunolabeling of the EGFPL10a transgene also enables visualization of the morphology and distribution of endothelial cells in the brain for anatomical studies. Hemizygous mice are viable and do not display and obvious gross physical or behavioral abnormalities.

Anti-GFP immunofluorescent labeling of central nervous system tissue reveals transgene expression in blood vessels (endothelial cells) throughout the brain and spinal cord. Endothelial cells in other tissues are also expected to be labeled, although this has not been checked. Native EGFP fluorescence (without immunohistological staining) is also visible in blood vessels from fixed brain tissue.

This bacTRAP transgenic strain, incorporating an EGFP-L10a ribosomal fusion protein targeted to the Abcb1a gene, enables cell labeling, sorting, and affinity purification of cell-specific polysomal RNAs associated with endothelial cells. Expression of endogenous Abcb1a is expected to be knocked out.
