Description

pOxr1-Cre mice express cre recombinase in the brain's superficial layer III of the medial entorhinal cortex (MEC). This mouse line has been used to manipulate neuronal circuits within the hippocampus and entorhinal cortex that play crucial roles in episodic memory and working memory.

Detailed description

Entorhinal cortex layer III neural input to the hippocampus, involving the Oxr1 (oxidation resistance 1) gene, is critical for temporal association memory.

These BAC transgenic mice express cre recombinase from the Oxr1 gene. When crossed with animals carrying a loxP-flanked region of interest, the floxed region is excised robustly and specifically in superficial layer III of the medial entorhinal cortex (MEC). This mouse line has been used to manipulate neuronal circuits within the hippocampus and entorhinal cortex that play crucial roles in episodic memory and working memory.

To selectively inhibit synaptic transmission at the MECIII to CA1/subiculum synapses via the use of the tetanus toxin light chain (TeTX), these pOxr1-Cre mice can be crossed with pαCaMKII-loxP-STOP-loxP-tTA mice (see Stock No. 010712) and tetO-teTX mice (see Stock No. 010713).

Development

BAC RP23-113L5, containing the complete coding sequences of the mouse Oxr1 gene (~80 kb of 5' sequences, all exons, and ~50 kb of 3' sequences), was modified to incorporate cre recombinase in the Oxr1 exon 2 translation initiation site. The construct was injected into fertilized C57BL/6NTac oocytes. This strain was maintained on a C57BL/6NTac genetic background by the donating laboratory. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

Molecular note: The cre recombinase sequence was inserted by BAC recombineering methods into the translation initiation site in exon 2 of the Oxr1 gene in a BAC construct which contained the promoter region, coding sequence and a 50 kb 3' downstream region. The final vector was injected into pronuclei of fertilized C57BL/6 eggs. 16 founders were obtained and crossed with lacZ reporter mice to identify one line which displayed robust recombination in superficial layer III the dorsal portion of the medial entorhinal cortex.
