RTP1,2DKO mice lack expression of both receptor transporter protein 1 (Rtp1) and receptor transporter protein 2 (Rtp2). These mice are useful for studying olfactory receptor trafficking.

Detailed description

RTP1,2DKO mice lack expression of both receptor transporter protein 1 (Rtp1) and receptor transporter protein 2 (Rtp2). These genes encode transmembrane proteins expressed in peripheral olfactory organs that enhance the trafficking of olfactory receptors to the cell surface. Mice that are heterozygous for both alleles are viable and fertile. The donating investigator reports that homozygous double knock-out (DKO) mice are viable but infertile.

RTP1,2DKO mice have olfactory receptor trafficking defects, a substantial reduction in the number of mature olfactory sensory neurons, and an overall diminished olfactory capacity. The expression of different olfactory receptors is altered compared to wildtype.

These mice were bred to B6;129P2-Olfr151^tm26Mom/MomJ (Stock No. 006676), co-expressing Olfr151 (also known as M71) and IRES-tauGFP in olfactory sensory neurons. GFP staining is observed in the dendrites of Olfr151 positive olfactory sensory neurons, however, immunostaining against OLFR151 is restricted to the cell body, due to inability of these neurons to traffic the receptor to the dendrite.

Development

A targeting vector was designed to replace the start codon of the receptor transporter protein 1 (Rtp1) gene with a loxP-flanked ACN cassette. The ACN cassette, containing the neomycin resistance gene and Cre recombinase gene under the control of angiotensin-converting enzyme promoter, is flanked by loxP sites. Cre-mediated recombination during spermatogenesis removes the cassette leaving one loxP site and results in disruption of the Rtp1 coding region. The construct was electroporated into 129S/SvEv x C57BL/6-derived EF-1 embryonic stem (ES) cells.

Another targeting vector was designed to replace the start codon of the receptor transporter protein 2 (Rtp2) gene with a puromycin resistance (pac) cassette. The construct was electroporated in the Rtp1 KO ES cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts and resulting chimeric RTP1,2DKO mice were bred with C57BL/6J mice for at least 10 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
