Description

These transgenic BAC mice express tamoxifen-inducible cre-ERT2 from the mouse Htr6 promoter. Expression occurs in Htr6-expressing cells, including a subclass of excitatory interneurons in the dorsal horn of the spinal cord.

Detailed description

These transgenic BAC mice express tamoxifen-inducible cre-ERT2 from the mouse Htr6 (5-hydroxytryptamine (serotonin) receptor 6; also called 5-HT6) promoter. Endogenous expression of Htr6 is not perturbed.

When crossed with mice carrying loxP-flanked sequences of interest, tamoxifen treatment of the progeny results in cre-mediated excision of the floxed region. This occurs in Htr6-expressing cells, including a subclass of excitatory interneurons in the dorsal horn of the spinal cord. The recombination pattern is in good agreement with the Htr6 in situ hybridization data available on the Allen Brain Atlas website. This mouse line would be a valuable tool for investigators who want to study Htr6-positive excitatory interneurons in the dorsal horn of the spinal cord.

Development

Expression of tamoxifen-inducible cre-ERT2 is driven from the mouse Htr6 promoter contained on BAC RP23-65B16. This BAC encodes 4 additional genes: Rnf186, Tmco4, Nbl1, and Minos1 (expression not tested). The modified BAC vector was introduced to C57BL/6J embryos. This strain was created and maintained on a C57BL/6J genetic background by the donating laboratory. This is founder line NIDA108.

May, 2018: A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. Several of the markers throughout the genome were segregating, suggesting the background incorporates genomic contributions from non-C57BL/6 sources.
