Description

The Prdx6 C47S knockin mice carry an amino acid substitution that abolishes peroxidase activity, but retains phospholipase A2 activity. This strain may be useful for studying lung cell recovery following oxidative stress.

Detailed description

PRDX6 (peroxiredoxin 6) is a bifunctional enzyme with phospholipase A2 and glutathione peroxidase activity and is involved in the repair of peroxidized phospholipids in lung cell membranes and the regulation of lung surfactant phospholipid turnover. The Prdx6 C47S knockin allele maintains phospholipase A₂ (PLA₂) activity, but lacks peroxide activity and is unable to reduce peroxides. Exposure to 100% oxygen (hypoxia) results in an intermediate degree of cell membrane repair (30-40%) as compared to complete recovery in wild-type mice and a markedly delayed recovery in Prdx6 null mice. Similar results are obtained using isolated perfused lungs and primary cell culture exposed to tert-butylhydroperoxide. Mice homozygous for the mutation are viable and fertile. This strain may be useful for studying lung cell recovery following oxidative stress.

Development

The targeting vector was designed to insert, by site-directed mutagenesis, a TGC to a TCC point mutation followed by a FRT-flanked neomycin cassette into exon 2. The mutation results in an alteration of the corresponding amino acid from cysteine to serine (C47S) at position 47. The construct was electroporated into EAP6 embryonic stem (ES) cells derived from C57BL/6 mice. Correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric animals were crossed to C57BL/6N females. Mice were crossed to B6.129S4- Gt(ROSA)26Sor^tm1(FLP1)Dym/RainJ to remove the neomycin cassette. The resulting progeny were crossed to remove the FLP1 allele. Upon arrival, mice were bred to C57BL/6J for at least 1 generation to establish the colony.
